l-tryptofaan is de bouwsteen van serotonine. Het is een essentieel aminozuur, hetgeen betekent dat het alleen via voeding in het lichaam wordt opgenomen; het lichaam maakt tryptofaan niet zelf aan. De methode van acute tryptofaandepletie omvat de inname van een mengsel dat ongeveer 100-gram grote aminozuren bevat in een natuurlijke verhouding, maar geen tryptofaan (zie de tabel).
